Molecular Anti-TSG101 antibody

TSG101 (tumor susceptibility gene 101 protein) is essential for endosomal sorting, degradation of membrane receptors and the final steps of cytokinesis. It plays a crucial role in cell proliferation and cell survival. TSG101 has been identified as a candidate tumor suppressor gene and belongs to the family of ubiquitin-conjugating enzymes. TSG101 is an exosome marker. This protein has 2 isoforms produced by alternative splicing with molecular mass 44 and 32 kDa.

The antibody recognizes the exosomal marker TSG101 as claimed. We tested it by Western blot in our preparations of isolated murine brain exosomes and it gave the signal at the correct molecular weight in the exosome-enriched fractions, so it appears to be specific. However, here we report some disadvantages that we encountered using this antibody (see attached file):1.

There is strong cross-reactivity of the antibody with the molecular weight marker we use (The Bio-Rad Dual Color Standard, catalog number 1610374)2. The signal to noise ratio is a bit low (the background noise coming from the membrane is quite high).

Positive control

This antibody gave a positive signal in the following whole cell lysates: HeLa (Human epithelial carcinoma cell line) A431 (Human epithelial carcinoma cell line) Jurkat (Human T cell lymphoblast-like cell line) HEK293 (Human embryonic kidney cell line) NIH 3T3 (Mouse embryonic fibroblast cell line) PC12 (Rat adrenal pheochromocytoma cell line) This antibody also gave a positive signal in human placenta tissue sections.
